  1.1.1. KRF (Korea Research Foundation) Research Project
  The Institute received funds from the Korea Research Foundation in October 2001 as a new government research project, which is a 6-year program. The grant consisted of $300,000 from the government and $209,000 from Kyung Hee University in 2004. The title of this project is ¡®Research of East-West medical treatment to strengthen the international competitiveness of Korean Oriental medicine¡¯, and the principle investigator of this project was professor Young-Seol Kim, former director of this institute.
Purpose of Research : The university chooses ¡®research of East-West medical treatment to strengthen the international competitiveness of Korean Oriental medicine,?? as the research subject of the institute, subdivides as the promotion domain ??globalization and strengthening of the competitiveness of Korean Oriental medicine,?? and emphasizes ¨ç constructing a collaborative research system of Eastern medicine and Western medicine, ¨è globalizing the resources of Korean Oriental medicine, and ¨é informationalizing the knowledge of Korean Oriental medicine as the main themes.

  The institute carried out research project for the Korea Science and Engineering foundation for the year 2004. The contents and result of the project are as follows. This project was carried out for the second year of the study period and terminated because of resignation of the PI. The fund of the project was $34,000, for one year.
Title : The study of the genetic polymorphism associated with rheumatoid arthritis by Sasang Constitution

Purpose of Research : In order to examine the genetic polymorphism associated with rheumatoid arthritis by Sasang Constitution, this study was planned to detect the susceptibility of the patients diagnosed by rheumatoid arthritis to Sasang Constitution and to examine the risk factor such as life style and environmental stress in order to profoundly study the genetic polymorphism like metabolic gene correlated with the susceptibility to rheumatoid arthritis by specific Sasang Constitution. In preliminary, experiment, the genetic polymorphism of TNF-alpha (G308A), GSTM1, GSTT1 and CYP1A1 were analyzed in rheumatoid arthritis patients and controls.
Contents of Research : This study was planned to detect the susceptibility of the patients diagnosed by rheumatiode arthritis to Sasang Constitution and its association with specific genetic polymorphism.
In order to analyze the susceptibility of rheumatoid arthritis to Sasang Constitution, 147 rheumatoid arthritis patients and 147 matched controls were assessed with QSCC¥± question for Sasang Constitution. Then the frequencies of patients by Sasang Constitution were compared to those of the controls, which were statistically analyzed and adjusted by age, sex, smoking status and BMI.
On the basis of results obtained during the first year, specific constitutions which were expected to be related to rheumatoid arthritis were studied by lifestyle(smoking and overweight, e.g.) and the hazardous environment for health, in detail.
Also, the genetic polymorphism of TNF-¥á(G308), GSTM1, GSTT1 and CYP1A1 associated with rheumatoid arthritis according to Sasang Constitution are analyzed with RFLP-PCR in patients with rheumatoid arthritis and control.
Effectiveness of Research : During the first year, 147 rheumatoid arthritis patients and 147 matched controls were assessed with QSCC¥± question for Sasang Constitution. Soeumin was the most susceptible to rheumatoid arthritis compared to Taeumin or Soyangin. It was statistically significant after adjusting age, sex, smoking status, alchol intake, BMI, and economic status(Adjusted OR[95 3.01[1.47-6.15]). It is suggested that the specific Sasang Constitution associated with rheumatoid arthritis is used as basic data to prevent the risk factors for rheumatoid arthritis. During the second year, differential effect of passive smoking on the association between Sasang Constitution and rheumatoid arthritis risk was found. This study showed that the GSTM1 and CYP1A1 genetic polymorphisms have differential effects on the association of active and passive smoking with rheumatoid arthritis and the presence of the heterozyous or homozyous variant types of CYP1A1 (WM or M/M) appears to enhance the magnitude of the association between active/passive smoke exposure and rheumatoid arthritis. The genetic polymorphism of rheumatoid arthritis patients and controls, such as TNF-¥á(G308A), GSTM1, GSTT1 and CYP1A1, associated with the susceptibility to rheumatoid arthritis by Sasang Constitution was analyzed for a preliminary results. Differential effects of TNF-¥á(G308), GSTM1, GSTT1 and CYP1A1 genetic polymorphism on the association between rheumatoid arthritis risk and Sasang Constitution were found. It is suggested that the genetic polymorphism correlated with susceptibility to rheumatoid arthritis by specific Sasang Constitution used as its susceptibility marker and further as basic data to prevent the risk factors for rheumatoid arthritis. But larger studies will be needed to confirm these preliminary findings.

Title : Studies on the Standardization of Processed Herbal Medicine

Herbal medicines are generally not used in the natural form, but in the processed form in order to make sure of their safety, efficacy, stability and uniformity of quality. Therefore, many herbal medicines are processed by heating, steaming and mixed frying with honey, wine, vinegar and brine etc. in order to improve the efficacy, decrease toxicity and side effects, and change the propertyies have used in oriental medicine in Korea, China and Japan. As a study of manufacturing process standardization and regulation of processed herbal medicines, we have been analyzing the chemical constituents before and after processing.
Meanwhile, the standardization of processing methods are necessary for the establishment of the guidelines which can be used for the application of effective and safe processed goods. Accordingly, in this study, we planned that we should select Glycyrrhizae Radix, Evodiae Fructus and Rhei Rhizoma which are commonly used, and establish the standard processing methods of processing Glycyrrhizae Radix by stir-frying and mix-frying with honey, processed Evodiae Fructus by mix-frying with licoris decoction and brine respectively, and processing Rhei Rhoma by mix-frying with vinegar and wine respectively, and char-frying, and furthermore, bring up the physicochemical parameters for the standard establishment of processed goods. And also, we intended that we contribute to the improvement of national health and welfare by the supply of safe herbal medicines into clinical medicine, and this could be fulfilled by security of the control processed goods for quality evaluation and standard establishment of processed goods.
For the purpose of guidelines for processing of standardized products of them, physico-chemical examination such as the contents of loss on drying, water extract, diluter ethanol, ether, residue on ignition and residue on acid insoluble ignition were tested. Also, in order to evaluate the quality control of them quantitative determination of standard compound from each herbal medicines have conducted HPLC method was apply to the quantitative analysis of pre-processed and post-processed.
(1) In frying or frying with watering methods using heat, manufacturing standardization needs the maintenance of regular temperature and fixed timely heating by use of rotary roaster having definite speed for obtaining the regularly qualified processed goods.
(2) In progressing method using liquid supplementary material, for manufacturing of processed Glycyrrhizae Radix by mix-frying with honey, processed Evodiae Fructus by mix-frying with licoris decoction and brine respectively, and processed Rhei Rhoma by mix-frying with wine, liquid supplementary material should be used in proper concentration by appropriate dilution for sufficient absorption. And then, processed goods should be left for prescribed time in the airtight space for the purpose of uniform absorption.
(3) Processed Glycyrrhizae Radix by stir-frying and mix-frying with honey are progressed goods of Glycyrrhizae Radix, and their glycyrrhizin(standard component) becomes glycyrrhetic acid, which is artifact, by heat decomposition. So, this artifact can be identified as standard component, and be utilized for the qualification standard by its quantification.
(4) As a identification method for processed Evodiae Fructus by mix-frying with licoris decoction, the identification of glycyrrhizin through TLC method can be used by reason of the fact that Glycyrrhizae Radix should be used for liquid supplementary material.
(5) As a identification method for processed Evodiae Fructus by mix-frying with brine, the qualification method of chloride compound is able to apply, on account of the reason that salt(NaCl) is used for supplementary material.
(6) In the establishment of standard progressing method of Rhei Rhizoma, diarrhea inducing activity and large intestine transportation activity are recommended as the standard of processed Rhei Rhizoma by mix-frying with vinegar, wine and char-frying, in turn.
