  5.5. Government Research Project

The Institute had received funds from the 'Korea Science and Engineering Foundation' on September 2000 as a government research project, which is a 3-year program. The research result of first year is as follows;

Title : Effects of acupuncture on alterations of immune responses evoked by hypothalamus lesions
Primary Investigator : Byung-Il Mim, MD. PhD,
Department of Physiology, College of Medicine, Kyung Hee University

To investigate effects of acupuncture and moxibustion on modulation of immune responses induced by lateral hypothalmic lesions, We made electrical lesions bilaterally (2mA, 15sec) on the lateral parts (2.0mm posterior to the bregma, 2.0mm laterally to the midline of the brain (or to the bregma), and 8.4mm in depth from the surface of the skull) of the hypothalmus of Spraque-Dawley rats (250-300g, age 6-8 weeks). These operations were also performed on a sham operation group without making any lesion. Lesioned rats were divided into 3 groups, and each group consisted of 9 rats. The first lesioned group was treated with moxibustion once a day (0.5mg moxibustion, three times per treatment) in the holder. The second lesioned group was treated with acupuncture once a day (1Hz, 0.25ms, 1-5V, 20min) in the holder. The third lesioned group was untreated (with only holder stress). The sham operation group was also included in this experiment
without any treatment (only with holder stress). In order to investigate time- dependent immune responses, 3 rats from each group were sacrificed on the 3rd, 13th, and 23rd days of treatments. The NK cell activity of each group was measured using 51Cr release immunoassay. T and B cell proliferation were measured using 3H- thymidine incorporation method. In rats sacrificed on the 3rd, 23rd days, the difference in NK cell activity and T and B cell proliferation between 4 groups was not significant. In rats sacrificed on the 13th day, NK cell activity and T and B cell proliferation in the lesioned, untreated group were markedly lower than that of sham operation group.
The moxibustion or acupuncture treatments in lesioned group significantly increase NK cell activity and T and B cell proliferation compared with lesioned control group. These results strongly suggest that acupuncture and moxibustion boost immune responses, and this effect is not mediated by lateral part of hypothalamus.

  (1) Preventive effects of cervine horn extract on experimental osteoporosis in mice.

Postmenopausal estrogen deficiency will occur osteoporosis. It may one of the leading cause of senile diseases. In Korea, ERT (estrogen replacement therapy) has been established to postmenopusal osteoporosis since a decade. We investigated corvine horn extract(CHE) as a natural resource of osteoporosis prevention. We administered CHE to ovariectomized and osteoporotic mouse, and then analyzed by bone histomorphometry. Finally, we found CHE could prevent osteoporosis on estrogen deficient state, but not treated previously made osteoporosis.

Figure 1. control (left) mouse revealed osteoporosis 12 weeks passed after ovariectomy, but CHE administered (right) mouse showed with ovariectomy showed no osteoposis at all.

We wondered if GH could modulate or help inflammatory processes in systemic or local inflammation. Its role in inflammation is not clear, but it could act an immune enhancer. Also, in a clinical point of view, GH was regarded as an immune suppressor. In preliminary experiment within our Lab., GH suppressed interleulin(IL)-1 beta, which was one of preinflammatory cytokines. We want to investigate GH role within autoimmune processes. We made autoimmune inflammation with type II collagen, which would develop arthritis in mice. We called it collagen- induced arthritis (CIA) in mice. It had been regarded as an animal model of rheumatoid arthritis. We would check specific changes of some inflammatory cytokines with CIA animals. It might be helpful for knowing GH roles in autoimmune reactions.

Heavy inflammatory cells, Lymphocytes and histiocyte infiltrates around the destructed tissue. DC treated (right): Some periosteal reactions are shown. Inflammatory cells are not found, and lining synoviums are normal looking.

The various preadipocyte cell lines and primary preadipocyte cultures have been used in studying the adipogenesis and adipolysis in vitro. The adipose conversion of mouse 3T3-L1 cells and other preadipocyte cell lines such as Ob17 and 3T3-FA442A, are known as a valid model for the development of adipose tissue in vivo. In this process of the adipose conversion, the cells develop large changes in gene expression and protein composition. According to this known and unknown profound change, the preadipocytes undergo
differentiation or arrest differentiation. And also, after the full differentiation, the adipocytes maintain their round lipid-filled differentiated state or activate lipolytic pathway according to many intrinsic or extrinsic stimuli. Interestingly, the extensive studies have found that the GH increase IGF-1 production, stimulate cell proliferation and inhibit their differentiation in primary preadipocyte cell cultures, but in preadipocyte cell lines such as 3T3-F442A, 3T3-L1, Ob 1771, and Ob 17UT, GH inhibit mitosis, alternate cytoskeletal protein expression and express of adipocyte-specific genes, thus promote preadipocyte differentiation to mature lipid-filled adipocyte. In this process, many known and unknown genes have been thought regulated and expressed by GH in time dependent manner. Hansen LH et al. reported that in primary rat preadipocyte culture, GH treatment prevent down-regulation of Pref-1 (that was suppressed during adipocyte differentiation), thus this unsuppressed expression of Pref-1 by GH possibly mediated the inhibitory effects on preadipocyte differentiation. So, first I observed the changes of the expression of adipocyte differentiation markers such as Pref-1, ADD-1, PPARr, and aP2 when GH was added to culture media during 3T3-L1 differentiation period (GH was added from differentiation day 0 to day 7). And second, GH was added to full- differentiated 3T3-L1 adipocyte (GH was added at culture media from the day 7 to day 14), the changes of the expression of differentiation markers were observed.

Figure 3. ADD-1 gene expression along GH concentration and culture periods. ADD-1 gene will express late differentiation time and depend on GH concentration.

(4) We carried out experiment for the role of herb and acupuncture on cellular toxicity test and COX-2 expression.
(5) We examined analgesic and anti-inflammatory effects of selected 100 herbs and made several complex recipes with the conduction of toxicity test.
(6) We made neuropathic pain animal model with the establishment of analgesic effects of drugs and acupuncture. We investigated the neuroprotective effects of Woohwangchungshimhwan (éÚüÜôèãýü¯).

During our second year, we will synthesize the information of selected drugs attained within the first year and construct the fundamental of development of therapeutic drugs for pain.
In part 1, we will search potential herbs and complex recipes for the treatment of arthritis and pain with research for therapeutic mechanism.
In part 2, we will make new complex recipes composed of selected herbs and will carry out experiment for the development of drugs for oral administration on the treatment of arthritis and pain.
In part 3, we will develop the therapeutic method for arthritis and pain using herbal acupuncture.
In part 4, we will develop the patch for treating arthritis.
In part 5, we will conduct a toxicity test for the examined drugs from parts 1-4.

7. Evaluation

A new government research project funded by the 'Korea Research Foundation' was initiated in October 2001. In the year of 2001, our institute has strengthened its research capabilities with the positive financial support of Kyung Hee University and government. We will continue our efforts on 'Vision 2000 Project, Kyung Hee University'. The International Symposium was held in October 2001, we have been thinking that it was a great success. We also hope that it was a good opportunity to modernize traditional medicine, and to increase the cooperation between western and oriental medicine. We will do our best to carry out plenty of research results and to play the role of WHO collaborating centre substantially.
